We have received a 5-year Transformative RO1 award from the NIDDK to use chimeric mice with fully mature and functional human liver tissue to accomplish two goals. (i) Demonstrate that human pharmacogenetic analyses can be performed in these mice. We have recently shown that chimeric mice can be detect liver toxicities that would be missed with conventional rodent models. (ii) Develop methods that enable autologous human liver replacement. This could enable a complete paradigm-shift where ‘liver transplantation’ is performed using autologous adipocyte-derived stem cells without immunosuppression. We have demonstrated that iPS cells can also reconstitute human liver, and we have begun modeling human genetic liver diseases in these mice. We have established an off-site barrier facility that is solely dedicated to breeding and experimentation on these mice. State of the art methods for ultra-sound guided cell transplantation into mice, stem cell differentiation are utilized.
Applicant Requirements:
M.D. and/or Ph.D. in a biologically relevant area and experience with the
following:
1) In vivo characterization of mouse models and/or pharmacology modeling
2) A publication record demonstrating productive use of in vivo modeling, a strong interest in biomedical research, and a demonstrated ability to deal with complex biological problems.
3) Experience with stem or iPS cell generation and experimentation is desired.
